Malarone is a combination of two medicines atovaquone and proguanil hydrochloride. It acts as a blood schizonticide and also has activity against hepatic schizonts of P.falcirum

It is indicated for:

Prophylaxis of Plasmodium falciparum malaria.

Treatment of acute, uncomplicated Plasmodium falciparum malaria.

Because Malarone is effective against drug sensitive and drug resistant P. falciparum it is especially recommended for prophylaxis and treatment of P. falciparum malaria where the pathogen may be resistant to other antimalarials.

Posology and method of administration.

A daily dose should be swallowed whole with a meal to ensure maximum absorption. If the patient is unable to tolerate food, the malanil/malarone can be administered but the absorption of atovaquone will be significantly affected. In an event of vomitting within an hour of dosing, a repeat dose should be administered.

Profylaxis

Administration should be commenced 24 to 48 hours prior to entering a malaria-endemic area. This should be followed by a once-daily dose during the entire stay and to be extended by 7 days after leaving the area.

Dosage in Adults

One Malanil/Malarone tablet daily. Malanil/Malarone tablets are not recommended for malaria prophylaxis in persons under 40 kg bodyweight.

Malanil/Malarone paediatric tablets are recommended for malaria prophylaxis in persons weighing <40 kg
